Molecular characterization of germplasm of patchouli (Pogostemon sp.) by RAPD markers

Abstract

The patchouli (Pogostemon sp.) is a herbaceous plant belonging to the Lamiaceae family, whose essential oil is widely used in cosmetics and perfumery. The study aimed to evaluate the genetic variability of patchouli accesses the Active Germplasm Bank of UFS by RAPD markers. For molecular characterization were used accessions POG-001, POG-002, POG-006, POG-014, POG-015, POG-016, POG-017, POG-019, POG and POG-021-022. We collected young leaves of access to DNA extraction. Then we proceeded to the purification and amplification using oligonucleotides 11, and the products separated on a 1% agarose gel stained with ethidium bromide and visualized under UV light. The genetic similarity among accessions was calculated by Jaccard Similarity coefficient and dendrograms were performed using the method of grouping unweighted arithmetic average - UPGMA. Genetic analysis by RAPD markers enabled the separation of patchouli accessions into four groups. There was significant variability among accessions.
